摘要: Littleisknownabouttheprevalenceofdrug-resistantmutationsinHIV-1-positiveindividualsinSuzhou,China.Toelucidatethetransmitteddrugresistance(TDR)andacquireddrugresistancemutation(ADR)profiles,wecollectedbloodspecimensfrom127drug-naiveand117first-linedrugtreatedHIV-1-infectedindividualssampledfrom2014to2016inSuzhou.Wesuccessfullyamplifiedpo/fragmentsfrom100drug-naiveand20drug-treatedsamples.Wethendeterminedthedrugresistantmutationstoprotease(PR)andreverse-transcriptase(RT)inhibitorsaccordingtotheStanforddrugresistancedatabase.Overall,11and13individualshadtransmitted(drug-naivegroup)andacquired(treatedgroup)resistancemutations,respectively.Sixtransmitteddrugresistantmutationswerefound,includingtwomutations(L33FandL76V)intheproteaseregionandfour(K70N/EandV179D/E)intheRTregion.OnlyL76Vwasamajormutation,andK70N/EandV179D/Eareknowntocauselow-levelresistancetoRTinhibitors.All13treatedparticipantswhohadmajordrugresistancemutationsdemonstratedintermediatetohighresistancetoefavirenzandnevirapine,andsixhadatreatmentdurationoflessthanthreemonths.NomajormutationstoRTinhibitorswerefound,implyingthattheepidemicoftransmittedresistancemutationswasnotsignificantinthisarea.Ourresultssuggestthatmorefrequentvirusloadanddrugresistancemutationtestsshouldbeconductedforindividualsreceivingantiretroviraltreatment,especiallyfornewlytreatedpatients.OurresearchprovidesinsightsintotheoccurrenceofHIV-1drugresistanceinSuzhouandwillhelptooptimizethetreatmentstrategyforthispopulation. ...
