摘要: Asystematicapproachwasdevelopedtoinvestigatethestabilityofgentamicinsulfate(GS)andGS/poly(lacticco-glycolicacid)(PLGA)coatingsonhydroxyapatitesurfaces.Theinfluenceofenvironmentalfactors(light,humidity,oxidationandheat)upondegradationofthedruginthecoatingswasinvestigatedusingliquidchromatographywithevaporativelightscatteringdetectionandmassspectrometry.GScoatedrodswerefoundtobestableacrosstherangeofenvironmentsassessed,withonlyanoxidizingatmosphereresultinginsignificantchangestothegentamicincomposition.Incontrast,rodscoatedwithGS/PLGAweremoresensitivetostorageconditionswithcompositionalchangesbeingdetectedafterstorageat60°C,75%relativehumidityorexposuretolight.Theeffectofγ-irradiationonthecoatedrodswasalsoinvestigatedandfoundtohavenosignificanteffect.Finally,liquidchromatography–massspectrometryanalysisrevealedthatknowngentaminesC_1,C_1aandC_2werethemajordegradantsformed.Forceddegradationofgentamicincoatingsdidnotproduceanyunexpecteddegradantsorimpurities. ...
